Zebrafish (Danio rerio) Vascular Endothelial Growth Factor A (VEGF-A) is a member of the vascular endothelial growth factor (VEGF) family, which includes VEGF-A, VEGF-B, VEGF-C, VEGF-D, and placental growth factor (PlGF)—key regulators of angiogenesis, vascular permeability, and endothelial cell growth. In zebrafish, VEGF-A is produced by multiple cell types including endothelial cells, macrophages, fibroblasts, epithelial cells, and developing embryonic tissues, particularly in response to hypoxia, inflammation, and tissue injury. VEGF-A exerts its biological effects primarily through binding to VEGF receptor homologs such as VEGFR-2 (Kdr/Kdrl) on endothelial cells, activating intracellular signaling pathways including MAPK/ERK, PI3K/AKT, and PLCγ, which promote endothelial cell proliferation, migration, vascular permeability, and formation of new blood vessels. In zebrafish, VEGF-A plays essential roles in embryonic vascular development, organogenesis, wound healing, and tissue regeneration, including regeneration of fins, heart tissue, and other organs. Because zebrafish embryos are transparent and genetically tractable, VEGF-A signaling has been extensively studied using live imaging and genetic models to investigate angiogenesis, vascular patterning, and responses to hypoxia or injury. As a result, zebrafish VEGF-A serves as an important model for understanding vertebrate vascular development and angiogenic signaling pathways relevant to human cardiovascular disease, cancer, and regenerative medicine.
