Tumor necrosis factor alpha (TNFSF2) is a member of the TNF Superfamily. It is produced chiefly by activated macrophages, but it is produced also by a broad variety of cell types including lymphoid cells, mast cells, endothelial cells, cardiac myocytes, adipose tissue, fibroblasts, and neuronal tissue. The primary role of TNF alpha is in the regulation of immune cells. TNF alpha, being an endogenous pyrogen, is able to induce fever, to induce apoptotic cell death, to induce sepsis (through IL-1 & IL-6 production), to induce cachexia, induce inflammation, and to inhibit tumorigenesis and viral replication.

Kingfisher Biotech's Equine TNFα was determined to be biological active by an independent laboratory in Spain. The principle of the assay is that Equine TNFα should stimulate an increase in production of IL-6 and iNOS in horse adult mesenchymal cells in culture. The Equine TNFα protein was reconstituted in 1 ml of PBS + 0.1% BSA (bovine serum albumin). The horse adult mesenchymal cells were cultured in 48 well plates, in the presence of TNFα (diluted 1:100) at a final concentration of 25 ng/ml. The cultures were maintained for 12 hours at 37 degrees C. RT-PCR and quantitative PCR (real time PCR) were used to analyze in the supernants of the cultures for IL-6 and the iNOS production. Compared to control cells, the cells stimulated with Equine TNFα had significantly higher levels of IL-6 and iNOS, showing that the Equine TNFα is biologically active.

Recombinant Equine TNF-α Catalog Number RP0137E contains a single amino acid substitution compared to NCBI reference sequence XP_005603547.1 (Leu at position 229 in place of Phe). This amino acid substitution was corrected to match the published sequence. The corrected recombinant protein is available (Catalog Number RP0930E-005/RP0930E-025) (Phe present at position 229).
