  • Bovine Interleukin-1 Receptor Antagonist (IL-1Ra) is an anti-inflammatory cytokine produced by monocytes, macrophages, neutrophils, and other immune cells in cattle (Bos taurus) that functions as a natural regulator of IL-1-mediated inflammation. IL-1Ra competitively binds to the IL-1 receptor type 1 (IL-1R1) without triggering downstream signaling, thereby inhibiting the pro-inflammatory effects of IL-1α and IL-1β and limiting activation of NF-κB and MAPK pathways. In bovine health, IL-1Ra plays an important role in modulating inflammatory responses associated with economically significant conditions such as mastitis, bovine respiratory disease (BRD), metritis, lameness, and enteric infections, helping to balance pathogen clearance with prevention of excessive tissue damage. Elevated IL-1Ra expression in the mammary gland during intramammary infection reflects its role in controlling neutrophil-driven inflammation and supporting resolution of inflammation. As a biomarker of anti-inflammatory regulation, bovine IL-1Ra is valuable in studies of immune balance, vaccine responses, and disease progression. In veterinary and translational research, characterization of bovine IL-1Ra supports investigations into inflammasome regulation, cytokine network dynamics, and development of strategies to mitigate excessive inflammation while preserving protective immunity in large-animal systems relevant to both livestock health and comparative immunology.
