  • The Black Flying Fox Bat CCL3 ELISpot assay is designed to evaluate CCL3-secreting cells by sensitively detecting and quantifying low-frequency chemokine-producing cells at the single-cell level. CCL3, also known as macrophage inflammatory protein-1 alpha (MIP-1α), is a chemokine that plays an important role in recruiting and activating immune cells such as monocytes, macrophages, and T lymphocytes to sites of infection and inflammation. In bats, CCL3 contributes to regulation of innate and inflammatory immune responses involved in host defense against viral pathogens. The black flying fox bat (Pteropus alecto) is an important reservoir host for several emerging zoonotic viruses, including Hendra virus, Nipah virus, Australian bat lyssavirus, and other paramyxoviruses and coronaviruses. Measurement of CCL3 is particularly valuable in bat immunology research because studying immune signaling in these bats helps reveal mechanisms of viral tolerance and host-pathogen interactions that allow bats to harbor viruses without developing severe disease. This assay enables researchers to quantify CCL3-producing cells to better understand chemokine-mediated immune cell recruitment and inflammatory responses in bats. The Black Flying Fox Bat CCL3 ELISpot kit contains a Capture Antibody, Detection Antibody, and Standard Control sufficient for the development of up to two (2) ELISpot assays. The included antibodies have been validated for performance in an ELISpot assay using the provided Standard Control. Additional materials and reagents required to complete the assay are not included. Black Flying Fox Bat (Pteropus alecto) CCL3 (C-C motif chemokine ligand 3, also known as MIP-1α, macrophage inflammatory protein-1 alpha) is a pro-inflammatory chemokine involved in recruitment and activation of immune cells during innate immune responses in bats. CCL3 primarily signals through the CCR1 and CCR5 receptors, promoting chemotaxis of monocytes, macrophages, natural killer (NK) cells, and T lymphocytes to sites of infection or tissue injury. In P. alecto, a natural reservoir for several zoonotic viruses including Hendra virus, Nipah virus, and other paramyxoviruses, CCL3 likely contributes to early immune cell recruitment and coordination of antiviral inflammatory responses. Produced by activated macrophages, dendritic cells, and epithelial cells, CCL3 participates in cytokine and chemokine networks that regulate leukocyte trafficking and immune signaling during infection. Importantly, bats are known for their ability to mount antiviral immune responses while limiting excessive inflammation; therefore, regulation of chemokines such as CCL3 may contribute to balanced immune activation that controls viral replication without causing severe immunopathology.
