  • The Bovine CCL2 ELISpot assay is designed to evaluate CCL2-secreting cells by sensitively detecting and quantifying low-frequency chemokine-producing cells at the single-cell level. CCL2, also known as monocyte chemoattractant protein-1 (MCP-1), is a chemokine that plays a key role in recruiting monocytes, macrophages, and other immune cells to sites of inflammation through interaction with the CCR2 receptor. In cattle, CCL2 is involved in regulation of inflammatory responses associated with infectious disease, tissue injury, and immune-mediated processes. Measurement of CCL2 is particularly important in bovine research for studying host responses to pathogens such as Mycobacterium bovis, bovine respiratory pathogens, and mastitis-associated infections, as well as broader mechanisms of inflammation and immune regulation in livestock health. The antibodies in this set recognize CCL2 with homology across multiple bovine species, including Bos taurus (cattle), Bison bison bison (American buffalo), Bos mutus (wild yak), and Bos indicus (zebu), supporting applications across domestic livestock and related species. This assay enables researchers to quantify CCL2-producing cells to better understand chemokine-driven monocyte recruitment and inflammatory immune responses. The Bovine CCL2 ELISpot kit contains a Capture Antibody, Detection Antibody, and Standard Control sufficient for the development of up to two (2) ELISpot assays. The included antibodies have been validated for performance in an ELISpot assay using the provided Standard Control. Additional materials and reagents required to complete the assay are not included. Bovine CCL2 (C-C motif chemokine ligand 2, also known as monocyte chemoattractant protein-1, MCP-1) is a pro-inflammatory chemokine that plays a central role in recruitment and activation of monocytes, macrophages, and memory T cells during immune responses in cattle (Bos taurus). CCL2 signals primarily through the CCR2 receptor, triggering intracellular signaling pathways that promote leukocyte migration, cytokine production, and coordination of innate and adaptive immune responses at sites of infection or tissue injury. In bovine health, CCL2 is particularly important in inflammatory conditions such as mastitis, where it mediates recruitment of monocytes and macrophages to infected mammary tissue, as well as in respiratory diseases associated with the bovine respiratory disease complex (BRDC) and infections such as Mycobacterium bovis. While CCL2-driven immune cell recruitment supports pathogen clearance and tissue repair, excessive or prolonged expression can contribute to tissue damage and chronic inflammation. As a biomarker of monocyte/macrophage recruitment and inflammatory activation, bovine CCL2 is widely studied in investigations of host–pathogen interactions, immune responses to bacterial and viral infections, and vaccine efficacy.
