  • The Bovine CCL4 ELISpot assay is designed to evaluate CCL4-secreting cells by sensitively detecting and quantifying low-frequency chemokine-producing cells at the single-cell level. CCL4, also known as macrophage inflammatory protein-1 beta (MIP-1β), is a chemokine that plays an important role in recruiting and activating immune cells such as monocytes, macrophages, natural killer cells, and T lymphocytes to sites of infection and inflammation through interaction with receptors including CCR5. In cattle, CCL4 contributes to regulation of innate and inflammatory immune responses involved in host defense against bacterial, viral, and parasitic pathogens. Measurement of CCL4 is particularly important in bovine research for studying immune responses associated with bovine respiratory disease complex, mastitis, and other infectious or inflammatory conditions affecting livestock health. The antibodies in this set recognize CCL4 with homology across multiple bovine species, including cattle, zebu, wild yak, and American buffalo, supporting applications across domestic livestock and closely related species. This assay enables researchers to quantify CCL4-producing cells to better understand chemokine-mediated immune cell recruitment and inflammatory responses. The Bovine CCL4 ELISpot kit contains a Capture Antibody, Detection Antibody, and Standard Control sufficient for the development of up to two (2) ELISpot assays. The included antibodies have been validated for performance in an ELISpot assay using the provided Standard Control. Additional materials and reagents required to complete the assay are not included. Individual components may also be purchased separately. Made in the USA. For research use only.

Bovine CCL4 (C-C motif chemokine ligand 4, also known as MIP-1β, macrophage inflammatory protein-1 beta) is a pro-inflammatory chemokine that regulates recruitment and activation of immune cells during inflammatory and infectious responses in cattle (Bos taurus). CCL4 primarily signals through the CCR5 receptor, promoting chemotaxis of monocytes, macrophages, dendritic cells, natural killer (NK) cells, and activated T lymphocytes to sites of infection or tissue injury. In cattle, CCL4 is produced by activated macrophages, dendritic cells, epithelial cells, and lymphocytes in response to inflammatory stimuli such as microbial components and cytokines including TNF-α and IL-1β. CCL4-mediated immune cell recruitment plays an important role in host defense against pathogens involved in diseases such as bovine respiratory disease complex (BRDC), mastitis, and mycobacterial infections including Mycobacterium bovis. In these conditions, infiltration of monocytes and lymphocytes contributes to pathogen clearance and activation of adaptive immune responses. However, excessive or prolonged expression of CCL4 may contribute to inflammatory tissue damage and chronic inflammation. As a regulator of leukocyte trafficking and inflammatory signaling, bovine CCL4 is important for studies of host–pathogen interactions, inflammatory disease mechanisms, and vaccine responses aimed at improving immune protection and disease resistance in cattle.
