  • The Ferret IFN-γ ELISpot assay is widely used to evaluate ferret cell-mediated immune responses by sensitively detecting and quantifying low-frequency, IFN-γ-secreting cells at the single-cell level. Interferon-gamma (IFN-γ) is a key pro-inflammatory cytokine produced primarily by activated T cells and NK cells and plays a central role in antiviral immunity, defense against intracellular pathogens, and Th1-type immune responses in ferrets. IFN-γ measurement is particularly important in ferret research due to their widespread use as models for human respiratory diseases, including influenza and SARS-CoV-2, where robust cellular immunity is critical for viral clearance and vaccine evaluation. This assay answers critical questions about immune function in ferrets, including vaccine efficacy, infection-induced immunity, T cell activation, and immunomodulatory effects of therapeutics by measuring the frequency of antigen-specific interferon-gamma (IFN-γ)-producing cells. The Ferret IFN-γ ELISpot kit contains a Capture Antibody, Detection Antibody, and Standard Control sufficient for the development of up to two (2) ELISpot assays. The included antibodies have been validated for performance in an ELISpot assay using the provided Standard Control. Additional materials and reagents required to complete the assay are not included. Ferret Interferon-Gamma (IFN-γ) is a type II interferon produced primarily by activated CD4⁺ and CD8⁺ T lymphocytes and natural killer (NK) cells in ferrets (Mustela putorius furo), where it plays a central role in cell-mediated (Th1-type) immunity. IFN-γ signals through the IFN-γ receptor complex (IFNGR1/IFNGR2), activating JAK/STAT1 pathways and inducing genes involved in macrophage activation, enhanced antigen presentation via upregulation of MHC class I and II molecules, and promotion of cytotoxic T-cell responses. In ferret health and research, IFN-γ is particularly important in models of respiratory viral infections such as influenza virus and SARS-CoV-2, where Th1 responses contribute to viral clearance but may also influence pulmonary inflammation and immunopathology. IFN-γ is also relevant in studies of bacterial infections, vaccine-induced immunity, and T-cell–mediated protection.
