  • The Swine IFN-γ ELISpot assay is widely used to evaluate porcine cell-mediated immune responses by sensitively detecting and quantifying low-frequency, IFN-γ-secreting cells at the single-cell level. Interferon-gamma (IFN-γ) is a key pro-inflammatory cytokine produced primarily by activated T cells and NK cells and plays a central role in antiviral immunity, defense against intracellular pathogens, and Th1-type immune responses in pigs. IFN-γ measurement is particularly important in swine research for studying diseases such as porcine reproductive and respiratory syndrome (PRRS), swine influenza, and Mycobacterium infections, as well as for evaluating vaccine-induced cellular immunity in production and translational models. This assay answers critical questions about immune function in pigs, including vaccine efficacy, infection-induced immunity, T cell activation, and immunomodulatory effects of therapeutics by measuring the frequency of antigen-specific interferon-gamma (IFN-γ)-producing cells. The Swine IFN-γ ELISpot kit contains a Capture Antibody, Detection Antibody, and Standard Control sufficient for the development of up to two (2) ELISpot assays. The included antibodies have been validated for performance in an ELISpot assay using the provided Standard Control. Additional materials and reagents required to complete the assay are not included. Swine Interferon-γ (IFN-γ) is a type II interferon cytokine that plays a central role in cell-mediated immunity and regulation of antimicrobial and inflammatory responses in pigs (Sus scrofa). IFN-γ is produced primarily by activated T helper 1 (Th1) cells, cytotoxic T lymphocytes, and natural killer (NK) cells following stimulation by intracellular pathogens or immune signaling molecules such as IL-12 and IL-18. Swine IFN-γ binds to the interferon-γ receptor (IFNGR1/IFNGR2) expressed on many immune and non-immune cells, activating intracellular signaling through the JAK–STAT pathway, particularly STAT1, which induces the expression of numerous interferon-stimulated genes involved in antigen presentation, macrophage activation, and antimicrobial defense. In pigs, IFN-γ is critical for activation of macrophages, enhancement of MHC class I and II expression, and control of intracellular pathogens, including viruses, bacteria, and parasites. Swine IFN-γ is widely studied in veterinary immunology and translational research, particularly in porcine models of infectious disease; for example, IFN-γ responses are commonly measured in studies of African swine fever virus (ASFV) infection and vaccine development, where IFN-γ-producing T cells serve as an important indicator of protective cellular immune responses.
