  • The Zebrafish IFN-γ 1-1 ELISpot assay is designed to evaluate zebrafish cell-mediated immune responses by sensitively detecting and quantifying low-frequency, IFN-γ 1-1-secreting cells at the single-cell level. Interferon-gamma 1-1 (IFN-γ 1-1) is one of the zebrafish IFN-γ homologs and plays an important role in macrophage activation, host defense against intracellular pathogens, and regulation of Th1-like immune responses in teleost fish. Measurement of IFN-γ 1-1 is particularly valuable in zebrafish research, which is widely used for studying innate and adaptive immunity, infectious disease models, inflammation, and translational immunology due to conserved immune pathways between zebrafish and mammals. This assay enables researchers to quantify antigen-specific or stimulated IFN-γ 1-1-producing cells to assess immune activation and functional cellular responses. The Zebrafish IFN-γ 1-1 ELISpot kit contains a Capture Antibody, Detection Antibody, and Standard Control sufficient for the development of up to two (2) ELISpot assays. The included antibodies have been validated for performance in an ELISpot assay using the provided Standard Control. Additional materials and reagents required to complete the assay are not included. Zebrafish Interferon-γ (IFN-γ) is a type II interferon cytokine that plays an important role in cell-mediated immunity and regulation of antimicrobial and inflammatory responses in zebrafish (Danio rerio). IFN-γ is produced primarily by activated T lymphocytes and natural killer (NK)-like cells following stimulation by intracellular pathogens or immune signaling molecules such as IL-12 and IL-18. Zebrafish IFN-γ binds to interferon-γ receptors expressed on various immune and non-immune cells, activating intracellular signaling through the JAK–STAT pathway, particularly STAT1, which induces expression of interferon-stimulated genes involved in antigen presentation, macrophage activation, and antimicrobial defense. In zebrafish, IFN-γ contributes to activation of macrophages, enhancement of antigen presentation pathways, and control of intracellular pathogens including bacteria and parasites. Zebrafish IFN-γ is widely studied in aquatic immunology and vertebrate disease models; for example, zebrafish models of Mycobacterium marinum infection use IFN-γ expression as a key marker of host immune responses during granuloma formation and mycobacterial pathogenesis.
