  • The Ovine IL-21 (Interleukin-21) ELISA is designed for the quantitative measurement of IL-21 in cell culture supernatants from sheep and can also be used to measure IL-21 in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, cerebrospinal fluid, and tissue lysates from organs such as lymph nodes, spleen, lung, liver, and intestine, supporting a broad range of immunology, inflammation, and translational research applications. IL-21 is a cytokine primarily produced by CD4⁺ T cells, particularly T follicular helper (Tfh) and Th17 cells, and it plays a central role in modulating B cell differentiation, promoting antibody production, stimulating NK and CD8⁺ T cell responses, and regulating inflammatory pathways. In the ovine model, IL-21 is widely studied in the context of viral and bacterial infections, autoimmune disorders, and vaccination responses, where its expression reflects immune activation and adaptive immune regulation. Measurement of IL-21 provides valuable insight into immune cell function, disease progression, and the efficacy of immunomodulatory or vaccine interventions. Reported measurements of ovine IL-21 have detected basal serum and plasma levels of approximately 10-150 pg/mL, with elevated levels-often 200-1,200 pg/mL-following infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-21 in other sample types to ensure optimal sensitivity and accuracy. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to further adjust assay parameters based on their specific sample type and experimental requirements.   • Ovine IL-21 (Interleukin-21) is a cytokine of the common γ chain (γc) family that plays a central role in regulating adaptive immune responses in sheep (Ovis aries). IL-21 is produced primarily by activated CD4⁺ T cells, particularly T follicular helper (Tfh) cells and Th17 cells, following antigen stimulation. It signals through the IL-21 receptor (IL-21R) in association with the common γ chain, activating downstream JAK/STAT pathways-predominantly STAT3-to regulate gene expression in B cells, T cells, and natural killer (NK) cells. IL-21 promotes B cell proliferation, plasma cell differentiation, immunoglobulin class switching, and antibody production, while enhancing CD8⁺ T cell cytotoxic activity and NK cell function. In healthy sheep, basal IL-21 expression is generally low but increases during infection, vaccination, and immune activation. Ovine IL-21 has been implicated in immune responses to pathogens such as ovine lentivirus (maedi-visna), Mycobacterium avium subspecies paratuberculosis, gastrointestinal nematodes (e.g., Haemonchus contortus), and respiratory pathogens. In veterinary and agricultural research, ovine IL-21 serves as a biomarker of T cell-dependent B cell help and humoral immunity and is studied in the context of vaccine efficacy, host-pathogen interactions, and regulation of adaptive immune responses in small ruminant health and production systems.
