  • The Caprine IL-1β (Interleukin-1 Beta) ELISA is designed for the quantitative measurement of IL-1β in cell culture supernatants from goats and can also be used to measure IL-1β in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, cerebrospinal fluid, synovial fluid, and tissue lysates from organs such as lung, spleen, liver, kidney, and lymph nodes, supporting a broad range of immunology, inflammation, and translational research applications. IL-1β is a pro-inflammatory cytokine primarily produced by monocytes, macrophages, and dendritic cells, and it plays a central role in initiating inflammatory responses, activating immune cells, and mediating fever and tissue repair. In the caprine model, IL-1β is widely studied as a key biomarker for immune activation during bacterial and viral infections, mastitis, respiratory diseases, and inflammatory disorders, making it critical for assessing goat health, disease susceptibility, and immune competence. Measurement of IL-1β provides valuable insight into immune system activation, disease progression, and the efficacy of vaccines or immunomodulatory interventions. Reported measurements of caprine IL-1β have detected basal serum and plasma levels of approximately 5-150 pg/mL, with elevated levels-often 200-2,000 pg/mL-following infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-1β in other sample types to ensure optimal sensitivity and accuracy. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to further adjust assay parameters based on their specific sample type and experimental requirements.   • Caprine Interleukin-1 Beta (IL-1β) is a potent pro-inflammatory cytokine produced primarily by activated macrophages, monocytes, and dendritic cells in goats (Capra hircus) in response to microbial infection, tissue injury, or inflammatory stimuli. Synthesized as an inactive precursor (pro-IL-1β), it is processed into its active form by caspase-1 within the inflammasome complex and signals through the IL-1 receptor (IL-1R1), activating NF-κB and MAPK pathways that promote fever, leukocyte recruitment, endothelial activation, and acute-phase protein production. In caprine health, IL-1β plays a central role in inflammatory responses associated with economically significant diseases such as mastitis, caprine arthritis encephalitis (CAE), pneumonia, caseous lymphadenitis, enteric infections, and parasitic infestations, where it contributes to both pathogen clearance and inflammatory tissue damage. Elevated IL-1β expression in the mammary gland is linked to neutrophil recruitment and severity of intramammary infection.
