  • The Cynomolgus Monkey IL-6 (Interleukin-6) ELISA is designed for the quantitative measurement of IL-6 in cell culture supernatants from cynomolgus monkeys (Macaca fascicularis) and can also be used to measure IL-6 in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, cerebrospinal fluid, and tissue lysates from organs such as spleen, liver, lung, and lymph nodes. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-6 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-6 is a pleiotropic cytokine produced by monocytes, macrophages, dendritic cells, endothelial cells, and fibroblasts, and it plays a central role in acute phase responses, inflammation, immune regulation, and hematopoiesis. In the cynomolgus monkey animal model, IL-6 is widely studied in the context of infectious disease, inflammatory and autoimmune disorders, vaccine development, toxicology, and translational research. The cynomolgus monkey IL-6 protein is 100% homologous to IL-6 from Macaca mulatta (rhesus monkey) and Macaca nemestrina (pig-tailed macaque), supporting its use in comparative and cross-species non-human primate studies. Measurement of IL-6 provides valuable insight into immune activation, cytokine regulation, disease progression, and the efficacy of therapeutic or immunomodulatory interventions, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions and increasing to tens to hundreds of picograms per milliliter in serum, plasma, or tissue-derived samples during infection, inflammation, or immune stimulation, depending on sample type and experimental context. Cynomolgus Monkey (Macaca fascicularis) Interleukin-6 (IL-6) is a multifunctional pro-inflammatory cytokine and a central member of the IL-6 cytokine family—including IL-6, IL-11, IL-27, IL-31, leukemia inhibitory factor (LIF), oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), and cardiotrophin-like cytokine factor 1 (CLCF1)—which signal through receptor complexes containing the shared gp130 subunit to activate JAK/STAT and MAPK pathways. Due to the close genetic and immunological similarity between cynomolgus monkeys and humans, IL-6 in this species exhibits high structural and functional homology to human IL-6, making it highly relevant in translational research. In cynomolgus monkeys, IL-6 regulates acute-phase protein production (including C-reactive protein), B- and T-cell differentiation, hematopoiesis, fever, and systemic inflammatory responses during infection, tissue injury, and immune stimulation. Elevated IL-6 levels are observed in models of viral infections (such as dengue, influenza, and SARS-CoV-2), bacterial sepsis, autoimmune disease, and cytokine release syndromes, where IL-6 contributes to inflammation and disease severity. As a key biomarker of immune activation, cynomolgus IL-6 is widely used in preclinical safety, vaccine, and therapeutic studies, particularly in evaluating biologics targeting the IL-6/IL-6R pathway.
