  • The Bovine IFN-β (Interferon Beta) ELISA is designed for the quantitative measurement of bovine interferon beta in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IFN-β in additional biological sample types reported in the literature, including serum, plasma, whole blood-derived culture supernatants, peripheral blood mononuclear cell (PBMC) culture supernatants, tissue homogenates or lysates, bronchoalveolar lavage fluid (BALF), and milk, depending on the experimental model and disease context. This flexibility supports a broad range of immunology and translational research applications. Interferon beta (IFN-β) is a type I interferon that plays a pivotal role in the early innate immune response, particularly during viral infection and immune stimulation. IFN-β is often one of the first interferons produced following pathogen recognition and acts as a key regulator of interferon-stimulated gene (ISG) expression, antiviral defense mechanisms, and immune cell activation. Measurement of IFN-β is widely used to evaluate innate immune signaling, antiviral responses, inflammatory regulation, and vaccine-induced immune activation. In bovine and related species, IFN-β serves as an important biomarker of early immune activation and disease progression, supporting research in animal health, infectious disease, and comparative immunology. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Bovine Interferon-Beta (IFN-β) is a type I interferon produced primarily by virus-infected epithelial cells, fibroblasts, macrophages, and dendritic cells in cattle (Bos taurus), where it serves as a key early mediator of innate antiviral immunity. Following recognition of viral nucleic acids by pattern recognition receptors such as RIG-I–like receptors and the cGAS-STING pathway, bovine IFN-β is rapidly induced and signals through the type I interferon receptor complex (IFNAR1/IFNAR2), activating JAK/STAT pathways and driving expression of interferon-stimulated genes (ISGs) that inhibit viral replication, enhance antigen presentation, and promote natural killer (NK) cell and adaptive immune activation. IFN-β plays an important role in host defense against economically significant bovine viral pathogens including bovine viral diarrhea virus (BVDV), bovine herpesvirus-1 (IBR), bovine respiratory syncytial virus (BRSV), foot-and-mouth disease virus (FMDV), and other agents involved in the bovine respiratory disease (BRD) complex. Many of these viruses possess immune evasion strategies that suppress or delay IFN-β production, contributing to viral persistence and disease severity. As both a biomarker and functional mediator of antiviral responses, bovine IFN-β is central to vaccine evaluation, antiviral research, and studies of host–virus interactions in cattle, while also supporting comparative research into type I interferon biology in large-animal models relevant to human infectious disease.
