  • The Bovine IL-17A (Interleukin-17A) ELISA is designed for the quantitative measurement of bovine IL-17A in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-17A in additional biological sample types reported in the literature, including serum, plasma, whole blood, peripheral blood mononuclear cell (PBMC) culture supernatants, tissue homogenates or lysates, milk, colostrum, bronchoalveolar lavage fluid (BALF), and synovial fluid, depending on the experimental model and disease context. This flexibility supports a wide range of immunology and translational research applications. Interleukin-17A (IL-17A) is a pro-inflammatory cytokine primarily produced by Th17 cells, γδ T cells, and innate lymphoid cells. It plays a central role in host defense, neutrophil recruitment, and mucosal immunity, and is involved in the regulation of inflammatory responses and autoimmune signaling. Measurement of IL-17A is widely used to assess cell-mediated immunity, inflammatory processes, and immune responses to infection or vaccination in bovine and related species. IL-17A serves as an important biomarker of immune activation and disease progression, supporting studies in animal health, infectious disease, comparative immunology, and translational research. The bovine IL-17A protein is 100% homologous to Banteng (Bos javanicus) and Zebu (Bos indicus) IL-17A, making this ELISA well suited for studies involving these species and closely related animals where sequence identity supports cross-species detection. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Bovine IL-17A (Interleukin-17A) is a pro-inflammatory cytokine belonging to the IL-17 family that plays a critical role in host defense and inflammatory responses in cattle (Bos taurus). IL-17A is produced primarily by Th17 cells, as well as by γδ T cells—which are abundant in ruminants—and other innate lymphoid cell populations in response to microbial infection and inflammatory stimulation. It acts on epithelial, endothelial, and stromal cells to induce expression of pro-inflammatory cytokines, chemokines (such as CXCL8), and antimicrobial peptides, thereby promoting neutrophil recruitment and enhancing mucosal and barrier immunity. In healthy cattle, basal IL-17A expression is generally low, but levels increase during bacterial and fungal infections, mastitis, respiratory disease, enteric infections, and parasitic challenges. IL-17A has been implicated in immune responses to pathogens such as Mycobacterium bovis, bovine respiratory disease complex agents, and gastrointestinal nematodes. In veterinary and agricultural research, bovine IL-17A serves as an important biomarker of Th17-mediated immunity and neutrophilic inflammation, providing insight into host–pathogen interactions, vaccine responses, and inflammatory disease mechanisms in cattle.
