  • The Canine VEGF-A (Vascular Endothelial Growth Factor A) ELISA is designed for the quantitative measurement of canine VEGF-A in cell culture supernatants and can also be used to measure VEGF-A in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and conditioned media, supporting a wide range of immunology, angiogenesis, and translational research applications. VEGF-A is a critical growth factor primarily produced by endothelial cells, fibroblasts, and immune cells, and it plays a central role in angiogenesis, vascular permeability, endothelial cell proliferation, and tissue repair. In canine models, VEGF-A is widely studied in the context of tumor angiogenesis, wound healing, cardiovascular disease, and inflammatory conditions, where its expression reflects vascular remodeling, tissue regeneration, and pathophysiological angiogenic responses. Measurement of VEGF-A provides valuable insight into angiogenic signaling, disease progression, and therapeutic efficacy. Canine VEGF-A shares 100% sequence homology with Canis lupus dingo (dingo), Nyctereutes procyonoides (raccoon dog), Vulpes lagopus (Arctic fox), and Vulpes vulpes (red fox), supporting precise cross-species studies. Reported measurements of canine VEGF-A have detected basal serum and plasma levels of approximately 30-200 pg/mL, with elevated levels-often 400-1,800 pg/mL-following tissue injury, inflammation, or tumor induction, and measurable expression has also been observed in tissue lysates and conditioned media from VEGF-A-producing cells. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Canine Vascular Endothelial Growth Factor A (VEGF-A) is a member of the vascular endothelial growth factor (VEGF) family, which includes VEGF-A, VEGF-B, VEGF-C, VEGF-D, and placental growth factor (PlGF)—key regulators of angiogenesis, vascular development, and endothelial cell function. In dogs (Canis lupus familiaris), VEGF-A is produced by a variety of cell types including endothelial cells, macrophages, fibroblasts, epithelial cells, and tumor cells, particularly in response to hypoxia, inflammation, or tissue injury. VEGF-A exerts its biological effects primarily through binding to VEGF receptor-1 (VEGFR-1/Flt-1) and VEGF receptor-2 (VEGFR-2/KDR) on endothelial cells, activating intracellular signaling pathways such as MAPK/ERK, PI3K/AKT, and PLCγ, which promote endothelial cell proliferation, migration, vascular permeability, and formation of new blood vessels. In canine physiology, VEGF-A plays important roles in normal vascular development, wound healing, and tissue regeneration. Dysregulated VEGF-A expression is also associated with tumor angiogenesis in canine cancers, including hemangiosarcoma, mammary tumors, mast cell tumors, and osteosarcoma, where increased VEGF signaling supports tumor growth and metastasis by promoting blood vessel formation.
