  • The Ovine IL-17A (Interleukin-17A) ELISA is designed for the quantitative measurement of IL-17A in cell culture supernatants from sheep and can also be used to measure IL-17A in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, cerebrospinal fluid, synovial fluid, bronchoalveolar lavage fluid, and tissue lysates from organs such as lymph nodes, spleen, lung, and intestine, supporting a broad range of immunology, inflammation, and translational research applications. IL-17A is a pro-inflammatory cytokine primarily produced by Th17 cells, γδ T cells, and innate lymphoid cells, and it plays a central role in recruiting neutrophils, stimulating antimicrobial peptide production, and amplifying inflammatory responses. In the sheep model, IL-17A is widely studied in the context of bacterial and viral infections, autoimmune disorders, and mucosal immune responses, where its expression reflects immune activation and the development of adaptive immunity. Measurement of IL-17A provides valuable insight into immune regulation, inflammation, and the efficacy of immunomodulatory interventions. Reported measurements of ovine IL-17A have detected basal serum and plasma levels of approximately 15-200 pg/mL, with elevated levels-often 250-1,800 pg/mL-following infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements.   • Ovine IL-17A (Interleukin-17A) is a pro-inflammatory cytokine belonging to the IL-17 family that plays a key role in host defense and inflammatory responses in sheep (Ovis aries). IL-17A is produced primarily by Th17 CD4⁺ T cells, as well as by γδ T cells-which are prominent in ruminants-and other innate lymphoid cell populations in response to microbial infection and immune stimulation. It acts on epithelial, endothelial, and stromal cells to induce expression of pro-inflammatory cytokines (such as IL-6 and TNF-α), chemokines (including CXCL8), and antimicrobial peptides, thereby promoting neutrophil recruitment and enhancing mucosal and barrier immunity in respiratory, gastrointestinal, and mammary tissues. In healthy sheep, basal IL-17A expression is generally low, but levels increase during bacterial and fungal infections, mastitis, respiratory disease, and parasitic challenges, particularly gastrointestinal nematode infections such as Haemonchus contortus. In veterinary and agricultural research, ovine IL-17A serves as an important biomarker of Th17-mediated immune activation and neutrophilic inflammation, providing insight into host-pathogen interactions, vaccine responses, and immune regulation in small ruminant health and production systems.
