  • The Chicken IL-6 (Interleukin-6) ELISA is designed for the quantitative measurement of IL-6 in cell culture supernatants from chickens and can also be used to measure IL-6 in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, bronchoalveolar lavage fluid, and tissue lysates from organs such as spleen, lung, liver, and intestine, supporting a broad range of immunology, inflammation, and avian translational research applications. IL-6 is a pleiotropic cytokine primarily produced by macrophages, T cells, and endothelial cells, and it plays a central role in stimulating acute phase responses, promoting B cell differentiation, and regulating inflammation and immune signaling. In the chicken model, IL-6 is widely studied in the context of bacterial and viral infections, inflammatory diseases, and vaccination responses, where its expression reflects immune activation and inflammatory signaling. Measurement of IL-6 provides valuable insight into immune system activation, disease progression, and the efficacy of immunomodulatory or vaccine interventions. Reported measurements of chicken IL-6 have detected basal serum and plasma levels of approximately 10-300 pg/mL, with elevated levels-often 400-2,000 pg/mL-following infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-6 in other sample types to ensure optimal sensitivity and accuracy. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to further adjust assay parameters based on their specific sample type and experimental requirements.   • Chicken Interleukin-6 (chIL-6) is a multifunctional pro-inflammatory cytokine that plays a central role in regulating innate and adaptive immune responses in avian species. Although the avian IL-6 cytokine system differs structurally from its mammalian counterparts, chicken IL-6 functionally parallels members of the mammalian IL-6 family by signaling through receptor complexes involving gp130 to activate JAK/STAT pathways and drive acute-phase responses, B-cell differentiation, and inflammatory gene expression. In chickens (Gallus gallus), IL-6 is rapidly induced in response to bacterial, viral, and parasitic infections-including avian influenza virus, Newcastle disease virus, infectious bursal disease virus, Salmonella, and Eimeria species-and contributes to fever, hepatic acute-phase protein production, and leukocyte activation. Dysregulated IL-6 expression is associated with excessive inflammation, reduced growth performance, and increased morbidity, making it an important biomarker for disease severity, vaccine response, and flock health monitoring in poultry production. In animal health and research settings, characterization of chicken IL-6 supports studies of host-pathogen interactions, mucosal immunity, vaccine development, and immune modulation strategies aimed at improving disease resistance, productivity, and welfare in commercial poultry systems.
