- The Ovine IL-6 (Interleukin-6) ELISA is designed for the quantitative measurement of IL-6 in cell culture supernatants from sheep (Ovis aries) and can also be used to measure IL-6 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lymph nodes, liver, and mammary gland. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-6 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-6 is a pleiotropic cytokine produced primarily by activated macrophages, T cells, and endothelial cells and plays a central role in inflammation, immune regulation, hematopoiesis, and acute phase responses. In the ovine animal model, IL-6 is widely studied in the context of infectious disease, mastitis, vaccination, and immunomodulatory research, where its expression reflects inflammatory and immune activation. Measurement of IL-6 provides valuable insight into immune responses, cytokine signaling, disease progression, and the efficacy of therapeutic or vaccine interventions, with reported levels in sheep typically detected at low nanogram-per-milliliter concentrations under basal conditions (~0.2-2 ng/mL in serum or plasma) and increasing during immune challenge, infection, or inflammatory stimulation, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant ovine IL-6 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Since ovine IL-6 is highly homologous to caprine (goat) IL-6, this assay is also useful for comparative studies across these closely related ruminant species. Ovine Interleukin-6 (IL-6) is a multifunctional pro-inflammatory cytokine and a central member of the IL-6 cytokine family—including IL-6, IL-11, IL-27, IL-31, leukemia inhibitory factor (LIF), oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), and cardiotrophin-like cytokine factor 1 (CLCF1)—which signal through receptor complexes containing the shared gp130 subunit to activate JAK/STAT and MAPK pathways. In sheep (Ovis aries), IL-6 plays a critical role in regulating acute-phase protein production (such as haptoglobin and serum amyloid A), leukocyte recruitment, B- and T-cell differentiation, fever induction, and systemic inflammatory responses during infection, tissue injury, and metabolic stress. Elevated IL-6 expression is associated with economically significant ovine diseases including mastitis, pneumonia, foot rot, caseous lymphadenitis, parasitic infestations, and viral infections such as ovine progressive pneumonia (maedi-visna), where it contributes to inflammation and disease severity. IL-6 also plays an important role in reproductive physiology and periparturient immune modulation. As a biomarker of immune activation, ovine IL-6 is valuable for herd health monitoring, vaccine evaluation, and assessment of inflammatory status.
