  • The Cynomolgus Monkey CCL2 (Chemokine (C-C motif) ligand 2, also known as MCP-1) ELISA is designed for the quantitative measurement of CCL2 in cell culture supernatants from cynomolgus monkeys (Macaca fascicularis) and can also be used to measure CCL2 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as lung, spleen, liver, and lymph nodes. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring CCL2 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. CCL2 is a chemokine produced primarily by monocytes, macrophages, endothelial cells, and fibroblasts and plays a central role in monocyte recruitment, inflammatory signaling, and immune cell trafficking. In the cynomolgus monkey animal model, CCL2 is widely studied in the context of infection, inflammation, cardiovascular research, and translational immunology, where its expression reflects chemokine-mediated immune activation. This assay can also measure CCL2 from Cercocebus atys (sooty mangabey), Chlorocebus sabaeus (green monkey), Macaca mulatta (rhesus monkey), Macaca nemestrina (pig-tailed macaque), Mandrillus leucophaeus (drill), Papio anubis (olive baboon), Pongo pygmaeus (Bornean orangutan), Rhinopithecus bieti (black snub-nosed monkey), and Rhinopithecus roxellana (golden snub-nosed monkey), highlighting its applicability across a wide range of Old World and great primate animal models due to the high sequence conservation of CCL2. Measurement of CCL2 provides valuable insight into immune responses, inflammatory pathways, disease progression, and the evaluation of therapeutic or vaccine interventions, with reported levels in primates typically detected at low picogram-per-milliliter concentrations under basal conditions (~10-150 pg/mL in serum or plasma) and increasing during immune challenge, infection, or inflammatory stimulation, depending on sample type and experimental context. Cynomolgus Monkey (Macaca fascicularis) CCL2 (C-C motif chemokine ligand 2, also known as monocyte chemoattractant protein-1, MCP-1) is a pro-inflammatory chemokine that plays a central role in recruitment and activation of monocytes, macrophages, dendritic cells, and memory T cells during immune responses. CCL2 primarily signals through the CCR2 receptor, activating intracellular pathways that promote leukocyte migration, cytokine production, and coordination of innate and adaptive immunity at sites of infection, inflammation, or tissue injury. Highly homologous to human CCL2, cynomolgus CCL2 is widely studied in nonhuman primate models of infectious and inflammatory diseases, including tuberculosis, dengue, influenza, and SARS-CoV-2, where it contributes to monocyte and macrophage recruitment to infected tissues and influences inflammatory disease progression.
