  • The Human IL-13 (Interleukin-13) ELISA is designed for the quantitative measurement of IL-13 in cell culture supernatants from humans (Homo sapiens) and can also be used to measure IL-13 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as lung, spleen, lymph nodes, and gastrointestinal tract. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-13 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-13 is a Th2-associated cytokine produced primarily by CD4⁺ T helper cells, innate lymphoid cells, mast cells, and eosinophils and plays a central role in type 2 immune responses, regulation of inflammation, mucus production, tissue remodeling, and fibrosis. In human studies and translational research, IL-13 is widely investigated in the context of allergic disease, asthma, parasitic infection, and mucosal immunity, where its expression reflects Th2-driven immune responses and host defense mechanisms. Measurement of IL-13 provides valuable insight into type 2 cytokine signaling, inflammatory pathways, disease progression, and the evaluation of immunomodulatory or therapeutic interventions, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions (~10-150 pg/mL in serum or plasma) and increasing during allergen exposure, infection, or inflammatory stimulation, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant human IL-13 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Human Interleukin-13 (IL-13) is a Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells, group 2 innate lymphoid cells (ILC2s), mast cells, and basophils, where it plays a central role in allergic inflammation, mucosal immunity, and tissue remodeling. IL-13 signals through receptor complexes composed of IL-13Rα1 and IL-4Rα, activating JAK/STAT6 pathways that regulate gene expression involved in IgE class switching, alternative (M2) macrophage activation, mucus production, epithelial barrier function, and suppression of certain pro-inflammatory cytokines. IL-13 is critically implicated in allergic diseases such as asthma, atopic dermatitis, chronic rhinosinusitis with nasal polyps, and eosinophilic esophagitis, where elevated IL-13 drives airway hyperresponsiveness, goblet cell hyperplasia, fibrosis, and tissue remodeling. Dysregulated IL-13 signaling also contributes to fibrotic diseases affecting the lung, liver, and other organs. As a key mediator of type 2 immunity, IL-13 has become a major therapeutic target, with monoclonal antibodies directed against IL-13 or the IL-4Rα receptor subunit demonstrating clinical benefit in allergic and inflammatory conditions. Consequently, human IL-13 is central to research on Th2-mediated inflammation, barrier immunity, fibrosis biology, and development of targeted cytokine-modulating therapies.
