  • The Mouse IFN-α (Interferon-alpha) ELISA is designed for the quantitative measurement of IFN-α in cell culture supernatants from mice (Mus musculus) and can also be used to measure IFN-α in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, liver, lung, lymph nodes, and bone marrow. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IFN-α in other sample types to ensure optimal sensitivity and accuracy, allowing adjustment of buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IFN-α is a type I interferon produced primarily by plasmacytoid dendritic cells, monocytes, and other immune cells and plays a central role in antiviral defense, innate immune activation, and modulation of adaptive immune responses. In the mouse animal model, IFN-α is widely studied in the context of viral infection, autoimmune disease, cancer immunotherapy, and innate immunity, where its expression reflects activation of type I interferon pathways and antiviral signaling. Measurement of mouse IFN-α provides valuable insight into immune system function, host defense mechanisms, disease progression, and the evaluation of immunomodulatory or therapeutic interventions, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions and increasing during infection, immune stimulation, or inflammatory challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant mouse IFN-α protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Mouse Interferon-Alpha (IFN-α) is a type I interferon comprising multiple closely related subtypes produced primarily by plasmacytoid dendritic cells and virus-infected cells in mice (Mus musculus), where it serves as a critical mediator of early innate antiviral immunity. Upon binding to the type I interferon receptor complex (IFNAR1/IFNAR2), murine IFN-α activates JAK/STAT signaling pathways, leading to induction of interferon-stimulated genes (ISGs) that inhibit viral replication, enhance antigen presentation, promote natural killer (NK) cell cytotoxicity, and shape adaptive immune responses. Mouse IFN-α is central to experimental models of viral infections including influenza virus, lymphocytic choriomeningitis virus (LCMV), murine cytomegalovirus (MCMV), West Nile virus, and SARS-CoV-2 (adapted strains), where timing and magnitude of interferon responses strongly influence viral control and immunopathology. Dysregulated or chronic IFN-α signaling has also been implicated in murine models of systemic lupus erythematosus, autoimmune disease, cancer, and interferonopathies. IFNAR-deficient and IFN-α transgenic mouse models have been instrumental in dissecting type I interferon biology, antiviral mechanisms, and immune-mediated tissue damage. As both a biomarker and functional regulator of host–pathogen interactions, mouse IFN-α remains central to preclinical research on antiviral therapies, vaccine development, cancer immunotherapy, and immune modulation strategies relevant to human disease.
