  • The Mouse IL-28B (IFN-λ3) ELISA is designed for the quantitative measurement of IL-28B in cell culture supernatants from mice (Mus musculus) and can also be used to measure IL-28B in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage fluid (BALF), intestinal tissue lysates, liver homogenates, and splenocyte culture supernatants, although minor protocol modifications may be required to optimize performance in different matrices. IL-28B is a member of the type III interferon (IFN-λ) family, signaling through the IL-28Rα (IFNLR1) and IL-10Rβ receptor complex, and plays a central role in antiviral immunity at epithelial barriers, particularly within the respiratory and gastrointestinal tracts. In the mouse animal model, IL-28B is widely studied in the context of viral infection (including influenza, respiratory viruses, and enteric pathogens), mucosal immunity, hepatic antiviral responses, and innate immune signaling, where it contributes to induction of interferon-stimulated genes (ISGs) while maintaining a more localized and tissue-restricted inflammatory profile compared to type I interferons. Measurement of mouse IL-28B provides valuable insight into epithelial antiviral defense mechanisms, cytokine signaling pathways, and host-pathogen interactions, with concentrations typically detected at low or undetectable levels under basal conditions and increasing rapidly following viral challenge or Toll-like receptor stimulation, depending on tissue source and experimental design. The kit contains a capture antibody, detection antibody, and recombinant mouse IL-28B protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Mouse interferon lambda 3 (IFN-λ3), also known as interleukin-28B (IL-28B), is a member of the type III interferon family and functions as a critical antiviral cytokine that signals through the IFNLR1 (IL-28Rα)/IL-10R2 receptor complex, activating JAK/STAT pathways and inducing interferon-stimulated genes (ISGs) that suppress viral replication primarily at epithelial barriers. In mice, IFN-λ3 plays a central role in mucosal antiviral immunity within the respiratory and gastrointestinal tracts, contributing to host defense against pathogens such as influenza virus, respiratory syncytial virus, norovirus, rotavirus, and SARS-like coronaviruses in experimental models. Unlike type I interferons, type III interferons act predominantly on epithelial cells and produce localized antiviral responses with limited systemic inflammation, making murine IFN-λ3 essential for studying barrier-specific immunity, viral transmission dynamics, and immune-mediated tissue protection.
