  • The Rabbit GM-CSF ELISA is designed for the quantitative measurement of granulocyte-macrophage colony-stimulating factor (GM-CSF) in cell culture supernatants from rabbits (Oryctolagus cuniculus) and can also be used to measure GM-CSF in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage fluid, peripheral blood leukocyte culture supernatants, and tissue lysates, with minor protocol modifications recommended to optimize assay performance across different matrices. GM-CSF is a hematopoietic growth factor and pro-inflammatory cytokine produced by activated T cells, macrophages, endothelial cells, fibroblasts, and other stromal cells in response to immune stimulation, and it functions through the GM-CSF receptor (CD116/CD131 complex) to promote the proliferation, differentiation, and activation of granulocytes and macrophages. In rabbit models, GM-CSF is particularly relevant in studies of inflammatory lung disease, wound healing, vaccine adjuvant responses, and host defense against bacterial and fungal infections, where it contributes to myeloid cell recruitment and activation. Measurement of rabbit GM-CSF provides valuable insight into myelopoiesis, innate immune activation, and cytokine-mediated inflammatory signaling, with basal concentrations typically low under normal physiological conditions and increasing following infection, immune stimulation, or tissue injury depending on experimental design and sample source. The ELISA kit contains a capture antibody, detection antibody, and recombinant rabbit GM-CSF protein standard validated to function together in an optimized ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Rabbit GM-CSF (Granulocyte–Macrophage Colony-Stimulating Factor, CSF2) is a hematopoietic cytokine belonging to the colony-stimulating factor family, which also includes G-CSF (CSF3) and M-CSF (CSF1) that regulate the development, proliferation, and activation of myeloid immune cells. In rabbits (Oryctolagus cuniculus), GM-CSF is produced by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to infection, inflammatory cytokines, or immune stimulation. Rabbit GM-CSF binds to the GM-CSF receptor (CSF2R) on hematopoietic progenitor cells and mature myeloid cells, activating intracellular signaling pathways such as JAK/STAT, MAPK/ERK, and PI3K–AKT, which promote the proliferation and differentiation of granulocytes, macrophages, and dendritic cells, while enhancing their phagocytic and antigen-presenting functions. In rabbit immunity, GM-CSF contributes to innate immune responses and inflammatory regulation, particularly through activation of neutrophils and macrophages during infection. Rabbits are also widely used as animal models for studying inflammatory and infectious diseases, and GM-CSF–mediated immune responses have been examined in rabbit models of bacterial pneumonia, where enhanced macrophage and neutrophil activation contributes to improved pathogen clearance and regulation of lung inflammation, providing insights relevant to host defense mechanisms and therapeutic strategies for respiratory infections.
