  • The Human CXCL7 (C-X-C motif chemokine ligand 7, also known as NAP-2 or PPBP) ELISA is designed for the quantitative measurement of CXCL7 in human cell culture supernatants and can also be used to measure CXCL7 in additional biological sample types reported in the literature, including serum, plasma, and platelet-rich plasma, with minor protocol modifications recommended to optimize performance across different matrices. The human CXCL7 protein sequence is identical to that of chimpanzee (Pan troglodytes), allowing this assay to be used for accurate detection in both species. CXCL7 is a platelet-derived CXC chemokine released during platelet activation and proteolytic processing of its precursor (pro-platelet basic protein), and it functions primarily through the CXCR2 receptor to promote neutrophil chemotaxis, activation, adhesion, and recruitment to sites of vascular injury or inflammation. In healthy individuals, circulating CXCL7 levels are influenced by platelet activation status and are typically low in carefully prepared plasma samples, but concentrations can increase significantly in serum or during conditions associated with platelet activation, thrombosis, cardiovascular disease, systemic inflammation, infection, or malignancy. In clinical and translational research, human CXCL7 serves as an important biomarker of platelet activation, thromboinflammatory responses, vascular injury, and neutrophil trafficking, with relevance in cardiovascular disease, autoimmune disorders, cancer, and inflammatory syndromes. The ELISA kit contains a capture antibody, detection antibody, and recombinant human CXCL7 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Human CXCL7 (C-X-C motif chemokine ligand 7), also known as NAP-2 (neutrophil-activating peptide 2) or PPBP (pro-platelet basic protein), is a platelet-derived chemokine belonging to the ELR⁺ CXC chemokine family, which includes CXCL1, CXCL2, CXCL3, and CXCL5 that regulate neutrophil recruitment during inflammatory responses. In humans (Homo sapiens), CXCL7 is produced primarily by platelets and megakaryocytes and is released during platelet activation following vascular injury, infection, or inflammatory signaling. CXCL7 signals mainly through the chemokine receptor CXCR2, which is expressed on neutrophils and other innate immune cells, promoting chemotaxis, activation, and migration of neutrophils to sites of tissue damage or infection. In human physiology, CXCL7 contributes to the coordination between hemostasis and immune responses, influencing platelet–leukocyte interactions, vascular inflammation, and tissue repair. Dysregulated CXCL7 activity has been associated with cardiovascular disease, thrombosis, inflammatory disorders, and cancer, where platelet-derived chemokines can affect angiogenesis, leukocyte recruitment, and tumor microenvironment signaling. As a result, human CXCL7 is widely studied in cardiovascular research, thrombosis biology, inflammation studies, and cancer immunology as a mediator of platelet-driven inflammatory and immune processes.
