  • The Human IL-6 (Interleukin-6) ELISA is designed for the quantitative measurement of IL-6 in human cell culture supernatants and can also be used to measure IL-6 in additional biological sample types reported in the literature, including serum, plasma, cerebrospinal fluid (CSF), bronchoalveolar lavage (BAL) fluid, synovial fluid, urine, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-6 is a pleiotropic pro-inflammatory cytokine produced by monocytes, macrophages, endothelial cells, fibroblasts, adipocytes, and other cell types in response to infection, tissue injury, and immune stimulation, and it plays a central role in the acute phase response, fever induction, B cell differentiation, T cell activation, and hepatic production of acute phase proteins such as CRP. IL-6 signals through the IL-6 receptor (IL-6R) and gp130 via both classical and trans-signaling pathways, contributing to immune regulation as well as chronic inflammation. In healthy individuals, circulating IL-6 levels are typically very low (often <1-5 pg/mL in serum or plasma), but concentrations can increase significantly-sometimes to tens, hundreds, or thousands of pg/mL-during acute infection, sepsis, autoimmune disease (e.g., rheumatoid arthritis), cytokine release syndrome, cardiovascular disease, chronic inflammatory disorders, and various malignancies, depending on disease severity and timing of sampling. Measurement of human IL-6 provides valuable insight into systemic inflammatory burden, cytokine storm dynamics, immune dysregulation, and therapeutic response in clinical and translational research settings. The ELISA kit contains a capture antibody, detection antibody, and recombinant human IL-6 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Human Interleukin-6 (IL-6) is a multifunctional pro-inflammatory cytokine and a central member of the IL-6 cytokine family—including IL-6, IL-11, IL-27, IL-31, leukemia inhibitory factor (LIF), oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), and cardiotrophin-like cytokine factor 1 (CLCF1)—all of which signal through receptor complexes containing the shared gp130 subunit to activate JAK/STAT and MAPK pathways. In humans, IL-6 is produced by macrophages, monocytes, endothelial cells, fibroblasts, and other cell types in response to infection, tissue injury, and immune stimulation, where it regulates acute-phase protein synthesis (including C-reactive protein and fibrinogen), B-cell maturation, T-cell differentiation (particularly Th17 development), hematopoiesis, and fever. Dysregulated IL-6 signaling is implicated in a wide range of pathological conditions, including rheumatoid arthritis, inflammatory bowel disease, systemic lupus erythematosus, cytokine release syndrome, sepsis, cardiovascular disease, metabolic syndrome, and numerous cancers such as multiple myeloma and lymphoma. Elevated IL-6 levels are closely associated with disease severity and poor clinical outcomes in inflammatory and infectious diseases, making it a critical biomarker and therapeutic target, as evidenced by the clinical use of IL-6 and IL-6 receptor inhibitors. In biomedical research and clinical medicine, human IL-6 and its related family members are central to studies of immune regulation, chronic inflammation, oncology, and targeted immunotherapy development.
