  • The Canine IL-6 (Interleukin-6) ELISA is designed for the quantitative measurement of IL-6 in dog (Canis lupus familiaris) cell culture supernatants and can also be used to measure IL-6 in additional biological sample types reported in the literature, including serum, plasma, cerebrospinal fluid (CSF), synovial fluid, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-6 is a pleiotropic pro-inflammatory cytokine produced by monocytes, macrophages, endothelial cells, fibroblasts, and other cell types in response to infection, tissue injury, and immune stimulation, and it plays a central role in the acute phase response, fever induction, B cell differentiation, T cell activation, and hepatic production of acute phase proteins. In healthy dogs, circulating IL-6 levels are typically low or near the limit of detection, but concentrations can increase significantly during systemic inflammation, sepsis, immune-mediated diseases, trauma, surgery, neoplasia, and infectious conditions such as parvovirus infection, ehrlichiosis, and leishmaniasis, depending on disease severity and timing of sampling. In veterinary and comparative medicine research, canine IL-6 is particularly relevant in naturally occurring disease models such as osteoarthritis, immune-mediated hemolytic anemia (IMHA), immune-mediated polyarthritis (IMPA), inflammatory bowel disease (IBD), chronic kidney disease, dilated cardiomyopathy, lymphoma, osteosarcoma, and mammary carcinoma, which parallel human inflammatory and oncologic disorders. Measurement of canine IL-6 provides valuable insight into systemic inflammatory burden, cytokine-driven disease progression, immune dysregulation, and therapeutic response in both veterinary and translational research settings. The ELISA kit contains a capture antibody, detection antibody, and recombinant canine IL-6 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Canine Interleukin-6 (IL-6) is a multifunctional pro-inflammatory cytokine and a central member of the IL-6 cytokine family—including IL-6, IL-11, IL-27, IL-31, leukemia inhibitory factor (LIF), oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), and cardiotrophin-like cytokine factor 1 (CLCF1)—all of which signal through receptor complexes utilizing the shared gp130 subunit to activate JAK/STAT and MAPK pathways. In dogs (Canis lupus familiaris), IL-6 regulates acute-phase protein production (such as C-reactive protein and serum amyloid A), B- and T-cell differentiation, hematopoiesis, and systemic inflammatory responses. Elevated IL-6 levels are associated with infectious diseases including canine parvovirus, distemper, and sepsis, as well as chronic inflammatory and immune-mediated disorders, osteoarthritis, and neoplasia. Importantly, several naturally occurring canine diseases serve as translational models for human conditions in which IL-6 plays a pathogenic role, including canine osteosarcoma (model for pediatric osteosarcoma), lymphoma (model for non-Hodgkin lymphoma), melanoma, atopic dermatitis (model for allergic skin disease), inflammatory bowel disease (model for human IBD), rheumatoid-like immune-mediated polyarthritis, and dilated cardiomyopathy.
