  • The Cynomolgus Monkey LIF (Leukemia Inhibitory Factor) ELISA is designed for the quantitative measurement of LIF in cell culture supernatants from Cynomolgus monkeys (Macaca fascicularis) and can also be used to measure LIF in additional biological sample types reported in the literature, including serum, plasma, cerebrospinal fluid (CSF), uterine lavage fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. The Cynomolgus monkey LIF protein sequence is identical to that of Rhesus monkey (Macaca mulatta), enabling this assay to be used for accurate detection in both species. LIF is a pleiotropic cytokine belonging to the IL-6 family and signals through the LIF receptor (LIFR) in complex with gp130 to regulate cell survival, differentiation, proliferation, and inflammatory responses. In non-human primates, LIF plays important roles in embryonic implantation and reproductive biology, hematopoiesis, neural development, tissue regeneration, and modulation of immune responses during infection and inflammation. In healthy Cynomolgus monkeys, circulating LIF levels are typically low or near the limit of detection, but concentrations may increase during inflammatory stimulation, tissue injury, reproductive cycling, or experimental immune challenge, depending on physiological and pathological conditions. Measurement of Cynomolgus monkey LIF provides valuable insight into cytokine-mediated regulation of development, immune balance, reproductive physiology, and tissue repair in preclinical safety, efficacy, and translational research studies. The ELISA kit contains a capture antibody, detection antibody, and recombinant Cynomolgus monkey LIF protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Cynomolgus Monkey (Macaca fascicularis) Leukemia Inhibitory Factor (LIF) is a multifunctional cytokine belonging to the IL-6 family—which includes IL-6, IL-11, oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), cardiotrophin-like cytokine factor 1 (CLCF1), and IL-27—and signals through receptor complexes containing the shared gp130 subunit. Highly homologous to human LIF, cynomolgus LIF binds to the LIF receptor (LIFR) in association with gp130, activating JAK/STAT3, MAPK, and PI3K signaling pathways that regulate cell survival, differentiation, immune modulation, and tissue repair. In nonhuman primates, LIF plays important roles in reproductive biology, including endometrial receptivity and embryo implantation, as well as in hematopoiesis, neural cell maintenance, and stem cell regulation. LIF-mediated STAT3 signaling is also involved in modulation of inflammatory responses and tissue remodeling during infectious disease models (e.g., dengue, influenza, SARS-CoV-2) and in studies of fibrosis and cancer biology. Dysregulated LIF expression may contribute to tumor progression or fibrotic pathology, making it relevant in oncology and safety pharmacology research. Because cynomolgus monkeys are widely used in preclinical evaluation of biologics and immune-modulating therapies, characterization of LIF supports translational investigations into IL-6 family signaling, STAT3-targeted therapeutics, reproductive medicine, and inflammatory disease mechanisms relevant to human health.
