  • The Rabbit C3a ELISpot assay is designed to evaluate C3a-secreting or C3a-releasing cells by sensitively detecting and quantifying complement activation at the single-cell level. Complement component 3a (C3a) is an anaphylatoxin generated during activation of the complement cascade and plays a critical role in innate immunity by promoting inflammation, leukocyte recruitment, vascular permeability, and modulation of adaptive immune responses. Measurement of C3a is particularly important in rabbit research for studying complement activation in models of inflammation, infection, immune complex disease, and antibody-mediated immune responses. Rabbits are also widely used in immunology, cardiovascular, and ophthalmic research where complement-driven inflammation contributes to disease mechanisms. This assay enables researchers to quantify C3a-associated cellular responses to better understand complement-mediated immune activation and inflammatory signaling. The Rabbit C3a ELISpot kit contains a Capture Antibody, Detection Antibody, and Standard Control sufficient for the development of up to two (2) ELISpot assays. The included antibodies have been validated for performance in an ELISpot assay using the provided Standard Control. Additional materials and reagents required to complete the assay are not included. Rabbit C3a is a biologically active anaphylatoxin generated during activation of the complement cascade in rabbits (Oryctolagus cuniculus). It is produced by proteolytic cleavage of complement component C3 and functions as a potent mediator of innate immune and inflammatory responses. C3a binds to the C3a receptor (C3aR) expressed on neutrophils, monocytes, macrophages, mast cells, endothelial cells, and other immune and stromal cells, triggering chemotaxis, cytokine release, histamine release, increased vascular permeability, and amplification of inflammatory signaling. In healthy rabbits, circulating C3a levels are typically low but rise rapidly during complement activation induced by bacterial or viral infection, immune complex formation, endotoxemia, ischemia-reperfusion injury, or experimental immune challenge. In biomedical research, rabbit C3a is particularly relevant in models of sepsis, atherosclerosis, ocular inflammation, immune complex–mediated disease, and cardiovascular injury, where rabbits are commonly used due to their suitability for vascular and lipid metabolism studies. Measurement of rabbit C3a provides valuable insight into complement activation, acute inflammatory responses, and innate immune regulation in experimental disease models.
