  • The Human EGF (Epidermal Growth Factor) ELISA is designed for the quantitative measurement of EGF in cell culture supernatants from humans (Homo sapiens) and can also be used to measure EGF in a variety of biological sample types reported in the literature, including serum, plasma, urine, saliva, and tissue lysates from organs such as lung, liver, kidney, skin, and gastrointestinal tract. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring EGF in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. EGF is a growth factor produced primarily by platelets, epithelial cells, and macrophages and plays a central role in cell proliferation, differentiation, wound healing, tissue regeneration, and epithelial maintenance. In human and non-human primate animal models, EGF is widely investigated in the context of wound healing, tissue repair, oncology, developmental biology, and regenerative medicine, where its expression reflects tissue growth and repair processes. This assay can also measure EGF from Pan paniscus (pygmy chimpanzee), Pan troglodytes (chimpanzee), and Gorilla gorilla gorilla (western lowland gorilla), highlighting its utility for comparative and translational studies across closely related primate species due to the high sequence conservation of EGF. Measurement of EGF provides valuable insight into growth factor signaling, tissue regeneration, disease progression, and the evaluation of therapeutic interventions, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions (~50-300 pg/mL in serum or plasma) and increasing during wound healing, tissue injury, or pathophysiological conditions, depending on sample type and experimental context. Human EGF (Epidermal Growth Factor) is a growth factor peptide belonging to the EGF family of ligands, which also includes transforming growth factor-α (TGF-α), amphiregulin (AREG), heparin-binding EGF-like growth factor (HB-EGF), betacellulin (BTC), and epiregulin (EREG) that signal through the epidermal growth factor receptor (EGFR/ErbB1) to regulate cell proliferation, differentiation, and tissue repair. In humans (Homo sapiens), EGF is produced by epithelial cells, fibroblasts, macrophages, and glandular tissues, particularly the salivary glands and gastrointestinal epithelium, where it plays important roles in maintenance and regeneration of epithelial tissues, including the skin, cornea, and intestinal lining. Binding of EGF to EGFR activates intracellular signaling pathways such as MAPK/ERK, PI3K–AKT, and JAK/STAT, promoting cell proliferation, survival, migration, and tissue remodeling during development and after injury. In human health, EGF is critical for wound healing, epithelial barrier maintenance, and tissue regeneration, and dysregulation of EGF–EGFR signaling is strongly associated with cancer development and progression, particularly in lung, colorectal, and head and neck cancers. Consequently, the EGF–EGFR signaling axis is a major focus of biomedical research and targeted cancer therapy, with EGFR inhibitors widely used as therapeutic agents in oncology.
